Nu Instruments | Carbonate Δ⁴⁷ Clumped Isotope Measurement in Micro-Volume Mode

This application note explains how the Perspective IS and Nu Carb carbonate preparation unit measure carbonate Δ⁴⁷ in micro-volume mode. It covers sample size range, micro-volume handling, calibration and precision using carbonate standards.

'Micro-volume' depletion during an analysis of a 500 pg Carrara marble sample. Closely matched sample and reference beams deplete from 12.4 V to 6 V 44 signal.

The Perspective IS coupled to the Nu Carb carbonate preparation unit enables carbonate Δ⁴⁷ clumped isotope measurement across a wide sample-size range. The document reports routine Δ⁴⁷ measurement from 150 µg to 6 mg, with precision below 0.02‰ in micro-volume mode.

Small samples are expanded into matched sample and reference micro-volumes, allowing the gas beams to deplete at closely matched rates during acquisition. Larger samples are handled using the dual inlet bellows, giving the system flexibility across different carbonate sample masses.

Calibration used heated and equilibrated CO₂ standards to build a transfer function and transform measured values into the absolute reference frame. Carbonate standards were then analysed, showing precise and accurate Δ⁴⁷ measurement across the reported sample range.

Key Takeaways:

  • Supports carbonate Δ⁴⁷ measurement from 150 µg to 6 mg.
  • Micro-volume mode enables full Δ⁴⁷ analysis from smaller carbonate samples.
  • Calibration against gas and carbonate standards supports accurate clumped isotope results.
